Carbetocin

Short Description:

Carbetocin is a long-acting synthetic oxytocin analog and potent oxytocin receptor agonist that exhibits high uterotonic activity both in vitro and in vivo. In isolated human and animal uterine smooth muscle preparations, carbetocin produces sustained, dose-dependent contractions with greater duration of action than oxytocin due to its enhanced metabolic stability. Pharmacological studies demonstrate strong affinity for oxytocin receptors while showing minimal vasopressin-related activity at therapeutic concentrations. In vivo, carbetocin effectively increases uterine tone, contraction frequency, and contraction amplitude in multiple animal models and clinical settings. A single intravenous or intramuscular dose of 100 μg provides prolonged uterine stimulation, with biological activity lasting significantly longer than oxytocin, reducing the need for repeated dosing. Carbetocin has been extensively investigated for the prevention of postpartum hemorrhage following cesarean section and vaginal delivery, where it effectively decreases uterine atony and blood loss. Its favorable pharmacokinetic profile, prolonged uterotonic effect, and established efficacy make carbetocin a valuable research tool for studying oxytocin receptor signaling, uterine contractility, labor physiology, and postpartum hemorrhage management.
